Production of HPV-18 E6/E7 gene expressing transgenic mice

Abstract
In this study, we have attempted to establish HPV-18 URR-E6/E7 expressing transgenic mice and to characterize the promoter activity of the upstream regulatory region(URR) of HPV-18.
Out of 40 mice born, four were identified as transgenic mice by polymerise chain reaction(PCR). The RT PCR was performed to asses tissue-specificity of the transgene expression. From a variety of tissues including brain, pancreas, kidney, heart, lung, liver and cervix, transgene-specific RNA was present exclusively in the uterine cervix.
In conclusion, transgenic mice specifically expressing in the uterine cervix were succefully generated F1. Thus, they could be useful as a model for studying on the development of cervix neoplastic change through a series of distinctive histologic stage. Moreover, this animal model will allow preclinical testing of compounds designed to interfere with the action of the HPV oncogenes or other clinical aspects of the cancer phenotype.
